AT-2490 is a synthetic peptide and zonulin antagonist originally developed by Alba Therapeutics. It is designed to regulate the permeability of the intestinal epithelia by inhibiting the zonulin pathway, which is responsible for the reversible opening of tight junctions. By blocking the action of zonulin (pre-haptoglobin 2), AT-2490 aims to prevent the pathological increase in intestinal permeability, often referred to as "leaky gut," which is implicated in the pathogenesis of various autoimmune and inflammatory diseases. The drug was primarily investigated for its potential in treating celiac disease and preventing the onset of type 1 diabetes. While it shares a similar mechanism of action with the more clinically advanced candidate larazotide (AT-1001), AT-2490 was an earlier or alternative candidate in the Alba Therapeutics pipeline and is no longer in active development.
