AT-527 + cyclosporine is a combination of two pharmaceutical agents. AT-527 is a novel, orally administered guanosine nucleotide prodrug that acts as an inhibitor of the hepatitis C virus (HCV) NS5B polymerase. It has demonstrated potent in vitro antiviral activity against HCV, including difficult-to-treat genotypes, and does not require activation by CYP enzymes, which differentiates it from other guanosine analogs[6]. Cyclosporine is an immunosuppressant drug primarily used to prevent organ rejection in transplant recipients and to treat certain autoimmune diseases such as rheumatoid arthritis and psoriasis. Its mechanism involves inhibition of calcineurin through binding to cyclophilin, thereby blocking T cell activation by preventing dephosphorylation and nuclear translocation of NFAT (nuclear factor of activated T cells), leading to reduced cytokine production[1][2][3][4][5].
