Overview

AT04B is a peptide-based therapeutic vaccine designed to treat hypercholesterolemia by targeting the proprotein convertase subtilisin/kexin type 9 (PCSK9) enzyme. Developed using the proprietary AFFITOPE technology, AT04B consists of short, synthetic peptides that mimic a specific epitope of the PCSK9 protein. These peptides are conjugated to a carrier protein, keyhole limpet hemocyanin (KLH), and adsorbed to an aluminum hydroxide adjuvant to enhance immunogenicity. Upon subcutaneous administration, the vaccine induces the production of high-affinity antibodies that bind to circulating PCSK9, preventing it from mediating the degradation of low-density lipoprotein receptors (LDLR) in the liver. This mechanism increases the density of LDLR on the surface of hepatocytes, facilitating the clearance of LDL cholesterol from the plasma. Originally developed by the Austrian biotech company AFFiRiS AG, the PCSK9 vaccine program, including AT04B, was acquired by Frontier Biotechnologies in 2021 for further clinical development.
