Overview

AT1019 is an investigational oncolytic virus therapy based on a recombinant herpes simplex virus type 1 (oHSV-1) platform. It is engineered to selectively replicate within and lyse tumor cells, a process that releases tumor-associated antigens and promotes a pro-inflammatory tumor microenvironment. Additionally, AT1019 is designed to express therapeutic transgenes, specifically human interleukin-12 (IL-12) and human OX40 ligand (OX40L), which act to enhance T-cell activation and overcome immune suppression. Developed by Aetio Biotech, AT1019 is currently being evaluated in Phase I clinical trials, including investigator-initiated trials at The Affiliated Hospital of Xuzhou Medical University, for the treatment of advanced solid tumors. In these studies, it is administered via intratumoral injection, often in combination with stereotactic body radiation therapy (SBRT) and PD-1 checkpoint inhibitors to evaluate potential synergistic effects through immune activation and the abscopal effect.
