AT247 is a novel, ultra-rapid acting prandial insulin analogue under development by Arecor. It is a second-generation formulation of insulin aspart designed to accelerate absorption after subcutaneous injection, closely mimicking the physiological mealtime insulin response in people without diabetes. The formulation includes excipients that bind calcium ions and transiently disrupt calcium-dependent cell adhesion at the injection site via reversible interactions with the calcium-cadherin complex, increasing tissue permeability and enabling faster absorption. This results in an earlier onset of action and enhanced early glucose-lowering effect compared to current best-in-class insulins such as NovoRapid (insulin aspart) and Fiasp (faster insulin aspart). Clinical studies have shown that AT247 achieves superior pharmacokinetic/pharmacodynamic profiles with no significant safety concerns reported. Its rapid action profile supports its potential use in advanced diabetes management technologies like closed-loop artificial pancreas systems[1][2][3][4][5][6][7][8].
