AT501 is a next-generation switchable chimeric antigen receptor T-cell (CAR-T) therapy candidate developed by AbClon for the treatment of HER2-positive solid tumors, specifically ovarian cancer. It is built on AbClon's proprietary zCAR-T platform, which employs an Affibody-based switch molecule to modulate the activation and activity of the CAR-T cells. This switchable mechanism is designed to overcome challenges associated with conventional CAR-T therapies, such as off-tumor toxicity and cytokine release syndrome, by allowing clinicians to control the 'on/off' state of the cells through the administration of a specific switch protein. In preclinical ovarian cancer models, AT501 has demonstrated the ability to completely eradicate cancer cells with a single dose.
