ATAD3A short hairpin RNA is an RNA interference (RNAi) agent designed to silence the expression of the ATAD3A gene. ATAD3A is a mitochondrial membrane protein that plays a pivotal role in the structural and functional integrity of mitochondria-associated endoplasmic reticulum membranes (MAMs). These membranes are critical for regulating calcium signaling, lipid metabolism, and mitochondrial dynamics. In various malignancies, including bladder cancer, ATAD3A is frequently overexpressed and correlates with tumor progression, metastasis, and resistance to therapy. By targeting ATAD3A mRNA for degradation, this shRNA disrupts MAM stability, leading to mitochondrial dysfunction, impaired metabolic homeostasis, and the induction of apoptosis in cancer cells. It is primarily utilized in preclinical research as a potential gene therapy approach to inhibit tumor growth and sensitize cells to chemotherapy.
