Overview
Atamestane + toremifene is an investigational combination therapy consisting of atamestane, a steroidal aromatase inhibitor, and toremifene, a selective estrogen receptor modulator (SERM). Atamestane inhibits the aromatase enzyme, thereby blocking the conversion of androgens to estrogens and reducing estrogen levels in the body. Toremifene acts as an antagonist of the estrogen receptor, blocking both circulating and intracellular estrogens from binding to their receptors. The combination aims for complete estrogen blockade by targeting both estrogen production (via aromatase inhibition) and receptor-mediated signaling (via SERM antagonism). This regimen has been studied primarily in postmenopausal women with hormone receptor-positive advanced breast cancer[1][2][3][4].
