Overview

Atazanavir + rosiglitazone is a combination of two small molecule drugs with distinct mechanisms and indications. Atazanavir is an azapeptide HIV-1 protease inhibitor used in combination with other antiretrovirals for the treatment of HIV-1 infection. It works by selectively inhibiting the virus-specific processing of viral Gag and Gag-Pol polyproteins in HIV-1 infected cells, preventing the formation of mature virions[2][4]. Rosiglitazone is a thiazolidinedione class anti-diabetic agent indicated as an adjunct to diet and exercise to maintain glycemic control in type 2 diabetes mellitus. It acts as a highly selective agonist for peroxisome proliferator activated receptor gamma (PPARγ), enhancing tissue sensitivity to insulin and regulating genes involved in glucose production, transport, and utilization[1][3][6]. There are significant drug-drug interactions between these agents; coadministration may result in elevated plasma concentrations of rosiglitazone due to inhibition of CYP2C8 by atazanavir[5].
