ATB-352 is a hydrogen sulfide-releasing derivative of ketoprofen, developed as a next-generation nonsteroidal anti-inflammatory drug (NSAID) for the treatment of acute and severe pain. Unlike traditional ketoprofen, which is associated with significant gastrointestinal toxicity, ATB-352 has demonstrated gastrointestinal safety in preclinical studies due to its ability to release small amounts of hydrogen sulfide that protect the GI tract lining. The drug acts primarily by inhibiting cyclooxygenase (COX) enzymes, thereby suppressing prostaglandin synthesis and reducing pain and inflammation. It also inhibits platelet aggregation. Importantly, preclinical data indicate that ATB-352 does not bind to opioid receptors and shows no addiction potential. The developer is pursuing both intravenous and oral formulations for perioperative pain management as an alternative to opioids[2][5][7].
