Overview

ATDC_Nantes is an autologous cell therapy consisting of tolerogenic dendritic cells (tDCs) developed by the University Hospital of Nantes (CHU de Nantes). These cells are generated *ex vivo* from a patient's own monocytes, which are harvested via leukapheresis and cultured with specific cytokines and pharmacological agents to adopt a stable, non-inflammatory, and immunosuppressive phenotype. Characterized by low expression of costimulatory molecules such as CD80 and CD86 and high production of anti-inflammatory cytokines like IL-10, these cells are designed to migrate to lymphoid organs upon re-infusion. Their primary mechanism involves inhibiting the activation of effector T-cells and promoting the expansion of regulatory T-cells (Tregs), thereby inducing long-term immune tolerance. This therapy is primarily investigated in the context of solid organ transplantation, specifically kidney transplantation, to prevent graft rejection while potentially reducing the requirement for lifelong systemic immunosuppression.
