Drug intelligence / Profile preview
Atebimetinib + anti-PD-1 is an investigational combination therapy comprising atebimetinib (IMM-1-104), an oral small-molecule Deep Cyclic Inhibitor of MEK, and an anti-PD-1 monoclonal antibody (such as cemiplimab or pembrolizumab). Developed by Immuneering Corporation, atebimetinib is designed to deeply suppress the MAPK pathway in a pulsatile fashion, allowing healthy cells to recover while preventing tumor resistance. Combining MEK inhibition with PD-1 checkpoint blockade aims to break tumor MAPK addiction and reverse MAPK-driven immune suppression, thereby enhancing anti-tumor immunity. The combination is being evaluated in clinical trials for the treatment of advanced solid tumors, including first-line RAS-mutant non-small cell lung cancer (NSCLC) and melanoma.
