Drug intelligence / Profile preview
IMM-1-104 + gemcitabine + Abraxane (atebimetinib + gemcitabine + paclitaxel protein-bound) is an investigational combination therapy being developed for the treatment of advanced solid tumors, particularly first-line metastatic pancreatic ductal adenocarcinoma (PDAC). The regimen combines atebimetinib (IMM-1-104), an oral, once-daily, clinical-stage dual MEK1/2 inhibitor designed as a 'deep cyclic inhibitor' to provide pulsatile inhibition of the MAPK pathway, with the standard-of-care chemotherapy backbone of gemcitabine (a nucleoside analog) and Abraxane (nab-paclitaxel, an albumin-bound nanoparticle formulation of the microtubule inhibitor paclitaxel). By combining pulsatile MEK inhibition with cytotoxic chemotherapy, the regimen aims to achieve deeper and more durable tumor responses in RAS-mutant and MAPK-driven cancers while minimizing overlapping toxicities. The combination is being evaluated in clinical trials, including the Phase 3 MAPKeeper 301 study.
