ateganosine + tislelizumab is a combination therapy being developed by MAIA Biotechnology for the treatment of colorectal cancer (CRC). It consists of ateganosine (THIO), a first-in-class small molecule telomere-targeting agent, and tislelizumab, a humanized monoclonal antibody targeting the PD-1 immune checkpoint. Ateganosine acts as a telomerase substrate that is preferentially incorporated into the telomeres of cancer cells, leading to telomere capping disruption, genomic instability, and immunogenic cell death. This process is intended to prime the tumor microenvironment, enhancing the efficacy of the subsequent PD-1 inhibition by tislelizumab, which restores T-cell activity against the cancer. The combination is currently being evaluated in Phase 2 clinical trials.
