Overview

Aterixen (XC221GI) is an oral small-molecule drug developed by Valenta Pharm for the treatment of mild to moderate COVID-19. It functions as a first-in-class inhibitor of the human RNA-binding protein HuR (Human antigen R, also known as ELAVL1). HuR plays a pivotal role in the post-transcriptional regulation of gene expression by binding to AU-rich elements in the 3' untranslated regions of various mRNAs, thereby increasing their stability and facilitating their transport from the nucleus to the cytoplasm. These target mRNAs include those encoding pro-inflammatory cytokines such as TNF-alpha, IL-1 beta, and IL-6, as well as components of the SARS-CoV-2 viral genome. By sequestering HuR within the nucleus, Aterixen suppresses the overproduction of cytokines—mitigating the risk of a cytokine storm—and inhibits viral replication. The drug was registered for use in Russia in March 2022.
