This drug is a combination of three monoclonal antibodies: **atezolizumab** (an anti-PD-L1 antibody), **bevacizumab** (an anti-VEGF antibody), and **IO-108** (a fully human IgG4 antibody targeting the myeloid checkpoint receptor LILRB2/ILT4). Atezolizumab blocks the interaction between PD-L1 and its receptors, enhancing T cell activation. Bevacizumab inhibits vascular endothelial growth factor (VEGF) to reduce tumor angiogenesis. IO-108 blocks LILRB2, a myeloid checkpoint receptor, disrupting cancer-associated myeloid immune suppression by blocking its interaction with ligands like HLA-G, ANGPTLs, SEMA4A, and CD1d. The combination is undergoing development as a first-line treatment for hepatocellular carcinoma (HCC) in a global Phase 1b/2 randomized study, under sponsorship by Roche and Immune-Onc Therapeutics[1][5].
