Overview

This combination therapy consists of atezolizumab (an anti-PD-L1 monoclonal antibody), carboplatin (a platinum-based chemotherapeutic agent), and nab-paclitaxel (albumin-bound paclitaxel, a taxane chemotherapy). The regimen works through multiple mechanisms: atezolizumab blocks the PD-L1 protein, enhancing T-cell immune responses against cancer cells; carboplatin forms DNA crosslinks, interfering with cell division; and nab-paclitaxel stabilizes microtubules, preventing cancer cell division. This combination was approved by the FDA in December 2019 for first-line treatment of metastatic non-squamous non-small cell lung cancer without EGFR or ALK genomic aberrations.
