Overview

This is an investigational combination regimen consisting of four agents: - **Atezolizumab** is a monoclonal antibody immune checkpoint inhibitor targeting programmed death-ligand 1 (PD-L1), enhancing anti-tumor immune responses. - **Ipatasertib** is a small molecule inhibitor of AKT, a key kinase in the PI3K/AKT signaling pathway, which plays a role in cell proliferation and survival. - **Nab-paclitaxel** (nanoparticle albumin-bound paclitaxel) is an albumin-bound formulation of the taxane chemotherapeutic agent paclitaxel, designed to improve delivery and reduce toxicity compared to conventional solvent-based paclitaxel. It disrupts microtubule function, inhibiting cell division[1][2]. - **Paclitaxel** is a standard taxane chemotherapy that also inhibits microtubule disassembly, leading to apoptosis in rapidly dividing cells. This combination brings together immunotherapy (atezolizumab), targeted therapy (ipatasertib), and two formulations of cytotoxic chemotherapy (nab-paclitaxel and paclitaxel). The rationale for combining these agents includes potential synergistic effects on tumor control by simultaneously targeting cancer cell survival pathways and enhancing immune-mediated tumor destruction. This regimen has been explored primarily in clinical trials for advanced or metastatic breast cancer.
