**Atezolizumab + lenvatinib** is an investigational combination treatment comprising two drugs: - **Atezolizumab**: a humanized monoclonal antibody (IgG1) targeting programmed death-ligand 1 (**PD-L1**), functioning as an **immune checkpoint inhibitor**. It blocks the interaction between PD-L1 and PD-1, restoring anti-tumor immune response. - **Lenvatinib**: an oral multikinase inhibitor (small molecule) targeting several receptor tyrosine kinases, notably **VEGFR**, **FGFR**, **PDGFRα**, **RET**, and **KIT**, thereby inhibiting angiogenesis and tumor cell proliferation. This combination is currently in clinical trials (Phase 3) for the treatment of **unresectable hepatocellular carcinoma (HCC)** in patients who have progressed after first-line therapy with atezolizumab plus bevacizumab. The regimen is postulated to offer immunomodulatory and anti-angiogenic effects, potentially overcoming tumor immune evasion and facilitating anti-tumor activity[5].
