atezolizumab + linagliptin

Atezolizumab + linagliptin is a combination of two pharmaceutical drugs investigated as a second-line treatment for locally advanced or metastatic gastric or gastroesophageal junction (GEJ) cancer. Atezolizumab (Tecentriq) is a monoclonal antibody that inhibits programmed death-ligand 1 (PD-L1), thereby enhancing anti-tumor immune responses. Linagliptin (Tradjenta) is a small molecule inhibitor of dipeptidyl peptidase-4 (DPP-4), typically used to treat type 2 diabetes, and hypothesized in this combination to increase anti-tumor immune activity by modulating immune cell recruitment and chemokine activity. The combination was studied in the phase 1b/2 MORPHEUS platform (MORPHEUS-GC, NCT03281369) but showed limited clinical benefit, with a 21.4% objective response rate in a small cohort[1][2][3].
