Overview

Atezolizumab + pirfenidone is an investigational combination therapy being studied for the treatment of advanced or recurrent non-small cell lung cancer (NSCLC). Atezolizumab is a monoclonal antibody that acts as an immune checkpoint inhibitor targeting programmed death-ligand 1 (PD-L1), thereby enhancing the immune system's ability to recognize and destroy cancer cells. Pirfenidone is a small molecule antifibrotic agent, primarily used in idiopathic pulmonary fibrosis, which inhibits transforming growth factor-beta (TGF-β)–mediated fibrogenesis and may modulate the tumor microenvironment. The rationale for combining these agents is to increase antitumor activity and reduce resistance to immunotherapy by targeting both immune evasion mechanisms and tumor-associated fibrosis[1][2][3][5][6].
