Overview

Atezolizumab + tocilizumab is an investigational combination of two monoclonal antibodies, each with distinct targets and mechanisms, studied in cancer immunotherapy. **Atezolizumab** is a humanized IgG1 monoclonal antibody that blocks programmed death-ligand 1 (PD-L1), interrupting its interaction with PD-1 and CD80 on T cells, and thus reactivating cytotoxic T-cell–mediated anti-tumor immunity. **Tocilizumab** is a humanized IgG1 monoclonal antibody that binds soluble and membrane-bound interleukin-6 receptor (IL-6R), inhibiting IL-6–mediated pro-inflammatory signaling implicated in tumor growth and immune modulation. The combination has been studied pre-surgery in localized prostate cancer and in recurrent glioblastoma, with the rationale to simultaneously relieve T-cell suppression by PD-L1 and block the tumor-promoting effects of IL-6 signaling, thereby enhancing anti-tumor immune responses[1][3][5].
