Drug intelligence / Profile preview
atezolizumab-IRDye800CW (Atezo800) is a near-infrared (NIR) fluorescently labeled monoclonal antibody conjugate developed for optical molecular imaging and fluorescence-guided surgery. It is prepared by conjugating the FDA-approved anti-PD-L1 monoclonal antibody atezolizumab with the near-infrared dye IRDye800CW NHS ester. The conjugate is designed to target and bind to programmed cell death 1 ligand 1 (PD-L1) on tumor cells, allowing real-time visualization of PD-L1 expression and tumor margins during surgical or endoscopic procedures. To overcome formulation challenges associated with free histidine in commercial atezolizumab, an automated manufacturing process utilizing tangential flow filtration (TFF) has been developed to pre-purify, conjugate, and purify the clinical-grade imaging agent.
