Overview

Atf3 siRNA is a small interfering RNA (siRNA) designed to silence the expression of Activating Transcription Factor 3 (ATF3). ATF3 is a member of the ATF/CREB family of transcription factors that is induced by various stress signals, including chronic ethanol exposure. In pancreatic β-cells, ATF3 acts as a negative regulator of glucose homeostasis by binding to the glucokinase (Gck) promoter, leading to Gck down-regulation and subsequent β-cell dysfunction. By utilizing RNA interference to degrade ATF3 mRNA, Atf3 siRNA restores Gck expression, ameliorates impaired glucose tolerance, and reduces insulin resistance. It is investigated as a potential therapeutic target for treating type 2 diabetes and alcohol-induced metabolic impairment.
