Overview

**ATFmmp14-IONP-DOX** is an investigational, uPAR-targeted theranostic iron oxide nanoparticle formulation carrying the anthracycline chemotherapy payload doxorubicin. Its targeting and stroma-penetrating ligand, ATFmmp14, combines the amino-terminal fragment of urokinase plasminogen activator with the catalytic domain of matrix metalloproteinase-14. The ATF domain directs the construct to urokinase plasminogen activator receptor-expressing tumor and stromal cells, while the MMP14 domain promotes extracellular-matrix remodeling to improve intratumoral penetration. Iron oxide enables MRI-guided tumor imaging, and released doxorubicin mediates cytotoxic antitumor activity. The construct has been evaluated preclinically, principally in pancreatic cancer models.
