Overview

ATG-021 is a novel "2+1" bispecific T-cell engager (TCE) antibody developed by Antengene for the treatment of multiple myeloma. It is designed to simultaneously target G-protein-coupled receptor class 5 member D (GPRC5D), which is highly and specifically expressed on malignant plasma cells, and the CD3 receptor on T cells. By bridging these two targets, ATG-021 induces T-cell activation and directs the cytotoxic killing of GPRC5D-expressing myeloma cells. Developed using Antengene's proprietary AnTenGager™ (TCE 2.0) platform, the molecule is engineered to maximize anti-tumor potency while minimizing the risk of cytokine release syndrome (CRS). Preclinical studies have demonstrated significant in vivo efficacy and a favorable safety profile compared to clinical-stage benchmarks.
