Overview

ATG-027 is an investigational **B7-H3 and PD-L1 bispecific antibody** discovered and developed by Antengene for cancer immunotherapy. It was described by the company as a potential first-in-class biologic for **hematologic malignancies and solid tumors**. The molecule was engineered by introducing a high-affinity PD-L1 single-chain variable fragment into a human IgG1 anti-B7-H3 antibody backbone, with a wild-type Fc region designed to preserve **ADCC** and **ADCP** effector function. Preclinical data presented at SITC 2022 indicated that ATG-027 can block the **PD-1 and PD-L1** interaction while also targeting **B7-H3**, promote T-cell activation, increase cytokine production, and show antitumor activity in vivo. Publicly available company materials placed the asset in preclinical development, while later database-style sources indicate the program appears to have been discontinued.
