Overview

ATG-110 is a novel "2+1" T cell engager (TCE) developed by Antengene. It is designed to target LY6G6D, a surface antigen expressed on certain tumor cells, and CD3, a component of the T cell receptor complex. The molecule has a unique structure in which the CD3 binding site is concealed until it binds to LY6G6D-expressing cells, reducing off-tumor activation and minimizing cytokine release syndrome (CRS). Preclinical studies have demonstrated potent T cell-mediated cytotoxicity against both high- and low-LY6G6D-expressing colorectal cancer cells with single-digit picomolar IC50 values. In vivo models showed complete tumor regression or significant suppression with minimal CRS risk. ATG-110 is being developed primarily for microsatellite stable (MSS) colorectal cancer, an area of high unmet need due to resistance to immune checkpoint inhibitors[1][4][5][7].
