Overview

ATG-201 is a novel bispecific T-cell engager (TCE) developed by Antengene Corporation. It is designed as a "2+1" CD19 x CD3 bispecific antibody, meaning it binds to both the B cell antigen CD19 and the T cell surface glycoprotein CD3. The drug’s mechanism leverages selective activation of T cells only in the presence of CD19-positive B cells, leading to potent and targeted depletion of B cells while minimizing cytokine release syndrome (CRS), a common toxicity with this class. Preclinical studies have shown that ATG-201 effectively depletes naïve and autoreactive B cells in vitro and in vivo, with significantly lower cytokine release compared to benchmark TCEs. It has demonstrated efficacy in models of lymphoma and autoimmune diseases such as systemic lupus erythematosus (SLE), showing deep depletion of B cells from blood, bone marrow, and spleen. The drug is being developed for the treatment of B cell malignancies (such as lymphomas) and various B cell-driven autoimmune diseases[1][2][4][5][6][7].
