Overview

ATG003 is a topical ophthalmic formulation of mecamylamine hydrochloride, a non-competitive, nonspecific nicotinic acetylcholine receptor (nAChR) antagonist. Developed by CoMentis, the drug was designed to treat neovascular (wet) age-related macular degeneration (nAMD) by targeting nAChRs located on retinal and choroidal endothelial cells. These receptors are involved in the pathways of angiogenesis and vascular permeability. ATG003 was formulated as an eye drop (0.3% and 1.0% concentrations) to provide local delivery to the posterior segment of the eye while minimizing the systemic side effects associated with oral mecamylamine, which was originally used as an antihypertensive. It was evaluated in Phase 2 clinical trials both as a monotherapy and as an adjunctive treatment to intravitreal anti-VEGF injections, with the therapeutic goal of reducing the overall injection burden for patients.
