Drug intelligence / Profile preview
ATG13 degraders are a class of targeted protein degraders (TPD) or small molecules designed to induce the degradation of autophagy-related protein 13 (ATG13). ATG13 is an essential adaptor protein and a core component of the ULK1 initiation complex, which is indispensable for autophagy induction. By promoting the degradation of ATG13, these compounds effectively inhibit autophagy—a key cell survival pathway that supports tumor growth, metabolism, and treatment resistance under stress. This therapeutic strategy is primarily being investigated for the treatment of KRAS-mutant cancers, such as non-small cell lung cancer (NSCLC) and pancreatic cancer. Sanford Burnham Prebys Medical Discovery Institute has developed high-throughput screening assays to identify these compounds, leading to the discovery of potent candidates like SBP-1750.
