Overview

ATH-063 is a first-in-class, oral, small molecule G9A inhibitor developed by Athos Therapeutics for the treatment of inflammatory bowel diseases (IBD), including ulcerative colitis and Crohn's disease. It acts as a genomic regulator targeting G9A enzymatic activity in human CD4 T cells and gastrointestinal epithelial cells. The drug suppresses pro-inflammatory responses by expanding and activating regulatory T cells (Tregs) and promotes direct mucosal healing through regulation of tight junction proteins. Preclinical data indicate that ATH-063 increases circulating Tregs and upregulates genes associated with anti-inflammatory activities. The compound was generated using Athos’ proprietary AI-driven drug discovery platform, aiming to provide precision therapeutics for immune-mediated diseases[1][2][4][5].
