Athebody DARPins for AAV vectors are engineered designed ankyrin repeat proteins (DARPins) developed using the proprietary Athebody platform by Athebio AG. These proteins are used as targeting ligands displayed on adeno-associated viral (AAV) vector capsids to enable highly specific gene delivery to cells expressing particular surface markers. The technology allows the generation of mono- or bispecific targeting domains, enabling precise cell-type targeting and improved safety profiles in gene therapy applications. The small size, high stability, reliable folding, and excellent target affinity of these DARPins make them particularly suitable for use with viral vector platforms such as AAVs. Proof-of-concept studies have demonstrated that bispecific DARPin-displaying AAVs can preferentially transduce cells expressing two specific markers (e.g., CD4 and CD32a), offering Boolean logic AND-gating at the cellular level and enhancing precision in therapeutic gene delivery[3][4]. The approach is being explored in partnership with companies like REGENXBIO to develop next-generation targeted gene therapies.
