Overview

The atherosclerosis-associated antigens vaccine is an experimental therapeutic vaccine candidate developed by researchers at the National Institute on Aging (NIA). The vaccine targets specific protein antigens identified from autoreactive B cells that reside within atherosclerotic plaques and human tibial arteries affected by peripheral artery disease. By inducing a robust, antigen-specific antibody response, the vaccine aims to clear pathogenic antigens before advanced disease develops, thereby reducing inflammation and plaque accumulation. In preclinical studies using ApoE-deficient mouse models, immunization with a combination of six high-priority antigens led to a significant reduction in arterial plaque accumulation compared to controls. This approach represents a novel immunotherapeutic strategy for the prevention and treatment of atherosclerosis and peripheral artery disease by targeting the underlying autoimmune components of arterial inflammation.
