Overview

**ATI-1428** is a fourth-generation, subnanomolar potent class II capsid assembly modulator (CAM) developed by Antios Therapeutics for chronic hepatitis B virus (HBV) infection. It features a novel ultra-potent mechanism that disrupts HBV pgRNA packaging and capsid assembly without accumulating empty capsids in hepatocytes, promoting a targeted antiviral immune response. Acquired by Antios in November 2021 from San Raffaele Hospital (OSR), Istituto Nazionale Genetica Molecolare (INGM), and IRBM/Promidis, it has demonstrated strong preclinical antiviral activity in vitro and in a transgenic mouse model of HBV, with favorable pharmacokinetics, positioning it as a potential component in functional cure regimens.[1][3][10]
