ATI-2138

ATI-2138 is a novel investigational oral small molecule covalent inhibitor developed by Aclaris Therapeutics. It acts as a dual inhibitor of interleukin-2-inducible T cell kinase (ITK), resting lymphocyte kinase, and Janus kinase 3 (JAK3). The drug demonstrates high potency and selectivity for both ITK and JAK3, with minimal activity against other related kinases. In preclinical studies, ATI-2138 showed disease-modifying activity in rodent models of arthritis and colitis. Early clinical trials in healthy human participants indicated that the drug is well tolerated with linear pharmacokinetics and dose-dependent inhibition of biomarkers linked to ITK and JAK3 activity. Its unique dual mechanism targets key inflammatory signaling pathways implicated in autoimmune and chronic inflammatory diseases.
