Overview

ATI-2173 is a liver-targeted, active site polymerase inhibitor nucleotide (ASPIN) developed by Antios Therapeutics for the treatment of chronic hepatitis B virus (HBV) infection. It is a phosphoramidate prodrug of clevudine monophosphate, designed to concentrate the active triphosphate metabolite specifically within the liver to inhibit HBV polymerase while minimizing systemic exposure. By targeting the active site of the viral polymerase, ATI-2173 aims to provide a potent and durable antiviral effect, potentially contributing to a functional cure when used in combination with other therapies such as RNAi therapeutics (e.g., AB-729) or capsid inhibitors (e.g., vebicorvir). However, the development of ATI-2173 was significantly impacted in May 2022 when the U.S. FDA placed a clinical hold on the program following a safety report, leading to the termination of major clinical collaborations.
