ATI-355 is a humanized monoclonal antibody developed by Novartis that targets the Nogo-A protein, an inhibitor of neurite outgrowth in the central nervous system. By binding to and neutralizing Nogo-A, ATI-355 promotes axonal regeneration and neuronal plasticity, which may improve functional recovery following acute spinal cord injury. The drug has been investigated primarily for intrathecal administration in patients with acute traumatic paraplegia and tetraplegia. Clinical studies have shown that ATI-355 is generally well tolerated, with most adverse events being mild and related to the underlying condition or route of administration rather than the drug itself. The mechanism of action involves steric hindrance of Nogo-A inhibitory domains as well as antibody-induced downregulation of Nogo-A expression[1][2][4][6].
