Overview

ATI-9494 is an investigational small molecule that acts as a potent and highly selective covalent inhibitor of Janus kinase 3 (JAK3). Developed by Aclaris Therapeutics using its proprietary KINect drug discovery platform, ATI-9494 is designed to treat autoimmune and inflammatory diseases by targeting the JAK-STAT signaling pathway. Unlike non-selective JAK inhibitors, ATI-9494 specifically targets the cysteine 909 residue in the ATP-binding pocket of JAK3, which is primarily expressed in hematopoietic cells and associates with the common gamma chain receptor. This selectivity is intended to minimize off-target effects associated with the inhibition of JAK1, JAK2, or TYK2, such as anemia or neutropenia. By blocking JAK3, the drug inhibits the signaling of key pro-inflammatory cytokines including IL-2, IL-4, IL-7, IL-9, IL-15, and IL-21, thereby modulating the activity of T cells and natural killer (NK) cells.
