Overview

Curve Therapeutics is developing a first-in-class small molecule ATIC inhibitor specifically for the treatment of MTAP-deleted cancers. ATIC (5-aminoimidazole-4-carboxamide ribonucleotide formyltransferase/inosine monophosphate cyclohydrolase) is a bifunctional enzyme that catalyzes the final two steps of de novo purine biosynthesis. In cancers with MTAP (methylthioadenosine phosphorylase) deletion, the purine salvage pathway is compromised, making the cells dependent on the de novo pathway and creating a synthetic lethal vulnerability. Unlike traditional antifolates, this compound inhibits ATIC by disrupting its homodimerization through a unique non-folate-dependent mechanism. Discovered using Curve's proprietary Microcycle platform, the program is currently in preclinical development for solid tumors including mesothelioma, pancreatic cancer, lung cancer, and glioblastoma.
