Overview

Aticaprant is a potent, selective, short-acting kappa opioid receptor (KOR) antagonist developed primarily for the treatment of major depressive disorder (MDD), especially as an adjunctive therapy in patients with inadequate response to standard antidepressants. It is administered orally and has demonstrated efficacy in reducing depressive symptoms and anhedonia. Aticaprant exhibits approximately 30-fold selectivity for KOR over mu-opioid and delta-opioid receptors, with minimal off-target effects at therapeutic doses. The drug has also been investigated for alcohol dependence, anxiety disorders, cocaine-related disorders, and smoking withdrawal but development was discontinued for these indications. Aticaprant was originally discovered by Eli Lilly and Company and further developed by Johnson & Johnson Innovative Medicine (Janssen) and Avalo Therapeutics[1][3][4][5][7].
