BMS-986489 is a fixed-dose combination (FDC) of atigotatug, a monoclonal antibody targeting T-cell immunoreceptor with Ig and ITIM domains (TIGIT), and nivolumab, a programmed death-1 (PD-1) immune checkpoint inhibitor. Developed by Bristol Myers Squibb, this combination aims to enhance anti-tumor immune responses by simultaneously blocking two distinct inhibitory pathways on T cells and natural killer (NK) cells. TIGIT is an inhibitory receptor expressed on various immune cells that competes with the activating receptor CD226 for binding to CD155 (PVR). By blocking TIGIT and PD-1, the combination seeks to overcome immune exhaustion and improve clinical outcomes in solid tumors, including extensive-stage small cell lung cancer (ES-SCLC), where it is being evaluated in Phase 3 trials (TIGOS) in combination with chemotherapy.
