Overview

Atilotrelvir (GST-HG171) is an oral small-molecule inhibitor of the SARS-CoV-2 3C-like protease (3CLpro), also known as the main protease (Mpro). Developed by Fujian Akeylink Biotechnology Co., Ltd., it is designed to treat mild to moderate COVID-19 by blocking the cleavage of viral polyproteins, a step essential for viral replication. In clinical use, atilotrelvir is typically administered in combination with ritonavir, which acts as a pharmacokinetic enhancer by inhibiting the CYP3A enzyme. The drug, marketed under the brand name Taizhongding in China, received conditional approval from the National Medical Products Administration (NMPA) in November 2023 based on clinical trials showing reduced recovery time and efficacy against multiple SARS-CoV-2 variants, including XBB.
