Overview

Atirmociclib + letrozole is an investigational combination therapy for hormone receptor-positive (HR+), human epidermal growth factor receptor 2-negative (HER2-) metastatic breast cancer. Atirmociclib (PF-07220060) is a next-generation, highly selective cyclin-dependent kinase 4 (CDK4) inhibitor developed to provide antitumor activity with reduced risk of neutropenia compared to less selective CDK4/6 inhibitors. Letrozole is a well-established aromatase inhibitor that suppresses estrogen production and is widely used in the treatment of HR+ breast cancer. The combination aims to enhance efficacy by targeting both cell cycle progression via CDK4 inhibition and estrogen-driven tumor growth via aromatase inhibition. Early clinical data indicate promising safety and preliminary efficacy as first-line therapy in HR+/HER2- metastatic breast cancer[1][3][7][8].
