ATL-024

ATL-024 is a novel antibody-drug conjugate (ADC) targeting the tumor-associated carbohydrate antigen CA242 (also known as CanAg), which is highly expressed in gastrointestinal malignancies such as colorectal, pancreatic, and gastric cancers. Developed by Antelope Therapeutics, ATL-024 consists of a humanized, Fc-silenced monoclonal antibody conjugated to the topoisomerase I inhibitor exatecan via a dipeptide cleavable linker, maintaining a drug-to-antibody ratio (DAR) of 4. The agent is designed to exploit the differential expression of CA242, which is virtually absent in normal tissues, thereby providing a favorable therapeutic index. Preclinical data demonstrate potent antitumor activity in various patient-derived xenograft (PDX) models and a promising safety profile in non-human primates. ATL-024 is currently in IND-enabling development for the treatment of CA242-expressing solid tumors.
