Overview

ATL844 is a potent and selective small molecule antagonist of the adenosine A2B receptor. It was primarily developed by Adenosine Therapeutics (later acquired by Clinical Data, Inc.) for the treatment of chronic inflammatory conditions, specifically asthma and chronic obstructive pulmonary disease (COPD). The mechanism involves blocking the A2B receptor on mast cells and other inflammatory cells, which prevents the release of mediators like histamine and cytokines that drive airway constriction and inflammation. Beyond respiratory indications, ATL844 has been investigated for its potential to improve glucose homeostasis and insulin sensitivity in the context of type 2 diabetes. Clinical development appears to have ceased following corporate acquisitions by Clinical Data and subsequently Forest Laboratories.
