Atlacabtagene autoleucel (Atla-cel) is a third-generation, autologous CD19-directed chimeric antigen receptor T-cell (CAR-T) therapy. Developed by BioOra Limited in partnership with Cincinnati Children's Hospital Medical Center, it is designed for the treatment of children and adolescents with relapsed or refractory B-cell acute lymphoblastic leukemia (B-ALL). The therapy involves the genetic modification of a patient's own T-cells to express a chimeric antigen receptor that specifically targets CD19, a protein found on the surface of B-cells. As a third-generation CAR-T therapy, it typically incorporates multiple costimulatory domains to enhance T-cell activation, persistence, and anti-tumor efficacy against malignant B-cells.
