ATM‐3507 (also known as Anisina) is a small molecule anti-tropomyosin agent developed as a first-in-class inhibitor targeting the Tpm3.1 isoform of tropomyosin, an actin-binding protein enriched in cancer cells. By disabling Tpm3.1-containing actin filaments, it disrupts the cytoskeleton and induces cytotoxicity in cancer cells. Preclinical studies demonstrated its efficacy against neuroblastoma and prostate cancer models both in vitro and in vivo, with notable synergy when combined with antimicrotubule agents such as vincristine or taxanes. The compound was selected for further development due to its potency and favorable drug-like properties but did not progress beyond preclinical stages[5][6][7].
