Overview

ATM-5292 is a first-in-class, orally bioavailable dual mRNA degrader and selective RNA-targeting splice modulator developed by Atomic AI using its proprietary PARSE™ machine learning platform. It is designed to target two key oncogenes, Forkhead Box M1 (FoxM1) and Myb (c-Myb), with equipotency. ATM-5292 potently modulates RNA splicing at low nanomolar concentrations, inducing the inclusion of a cryptic exon in both FoxM1 and Myb transcripts. This splicing alteration introduces a premature stop codon, triggering nonsense-mediated decay (NMD) and subsequently reducing the protein levels of both oncogenic targets. Preclinical studies have demonstrated that ATM-5292 exhibits robust anti-tumor activity and cytotoxicity across multiple cancer cell lines, including non-small cell lung cancer (NSCLC), breast cancer, colorectal cancer, and ovarian cancer, as well as in leukemia cell line-derived xenograft (CDX) models.
