ATN-101 (also known as ATG-101) is a first-in-class bispecific monoclonal antibody that simultaneously targets Programmed Cell Death 1 Ligand 1 (PD-L1) and 4-1BB (also known as CD137 or Tumor Necrosis Factor Receptor Superfamily Member 9). Developed by Antennova, a subsidiary of Antengene, the drug is designed to activate 4-1BB-mediated T-cell signaling only upon binding to PD-L1-expressing tumor cells. This conditional activation is intended to localize immune stimulation to the tumor microenvironment, thereby enhancing anti-tumor efficacy while minimizing the systemic toxicities typically associated with 4-1BB agonists. ATN-101 has received Orphan Drug Designation from the U.S. FDA for the treatment of pancreatic cancer and is currently being evaluated in Phase I clinical trials for advanced solid tumors, including colorectal cancer and pancreatic cancer.
